In an animation project, the first defined layer mask will also delete the corresponding original layer content?

I use layer masks in the layers of animation projects. My customary operation is to first create a new layer mask, then use the selection tool to draw a range on the mask, and use the delete operation to delete the areas on the mask. An area to mask the content on the original layer.
But recently I discovered that this operation will also delete the content of the original layer? ! Is this a bug?

After repeated attempts, I found that it is the same in all projects. As long as the mask is selected and deleted, the contents of the original layer will also be deleted!

I couldn't guess the meaning of your question from your words alone.
I imagine that in the animation function, masks are operated on a "cell layer" basis, but I'm not sure.
Can you post a screenshot of the entire operation screen, and in particular a screenshot that shows the relationship between the animation folder and the mask image, in the answer section of this question?

It's normal after testing. Maybe you selected the layer instead of the mask when selecting the layer?
